Pharm MCQs

Which of the following agents will bind to the purine base shown in the image and cause the changes observed?

a. Azathioprine
b. Vincristine
c. Cyclosporine
d. Cisplatin
e. Paclitaxel

Answer D
1. An alkylating antineoplastic agent is an alkylating agent used in cancer treatment that attaches an alkyl group (CnH2n+1) to DNA.
2. The alkyl group is attached to the guanine base of DNA, at the number 7 nitrogen atom of the purine ring.
3. Cisplatin is classified as an alkylating agent. Alkylating agents are most active in the resting phase of the cell. These drugs are cell cycle non-specific.
4. They stop tumor growth by cross linking guanine nucleobases in DNA double-helix strands, directly attacking DNA. This makes the strands unable to uncoil and separate. As this is necessary in DNA replication, the cells can no longer divide. These drugs act non-specifically.
